目前并没有哪一种粒径分析方法可以适用于所有的应用场景,在以上应用场景中,根据颗粒物性质、粒径范围等特征,广泛的使用了多种粒径分析方法,如沉降法、筛析法、影像分析法、X射线衰减法、电阻法、光散射法和光衍射法等[10],由此也有了很多依据各种原理生产的粒度测量仪器。其中基于显微镜、激光成像和光散射的粒径测量方法越来越广泛的应用于各个场景,具体的分类以及测量范围如图 1和图 2所示。本文作者将针对这3类方法的基本原理、适用范围、应用现状进行分析和比较。

显微镜法是一种简单、直观的测量方法,可以同时观察和测量颗粒的粒径、形状和表面形貌,对于非球形、不规则的颗粒物的测量具有独特的优势。它的缺点是计量数目有限、难以实现智能化的粒径分析。

普通光学显微镜法通过拍摄图像和长度换算来对颗粒粒径进行定值,将样品制备成玻片,由光学显微镜拍摄图像,根据显微镜放大倍数可以得到像素尺寸与实际长度的换算关系,统计颗粒占据的像素个数就可以计算出颗粒粒径[11]。此方法定值精准, 而且操作简单、直观,可以测量单个颗粒的粒度,测量范围为:0.8μm~150μm。但该方法只适合于粒度分布窄(粒径最大值比最小值小于10 ∶1)的样品。对于粒度分布宽的样品,粒径测量比较麻烦, 而且计量数目有限,往往缺乏代表性。
普通光学显微镜法广泛用于微米级粒子粒径的检测,如:食品存储中纳米凝胶乳液液滴聚集(7μm~90μm)的测量[12];二氧化硅乳液液滴(6μm~50μm)的剪切稳定性研究[13];海砂(70μm~600μm)-海水混凝土性能的研究[14];Fe和Si对Al-Mn合金晶粒(约几微米)生成的影响[15]等。

受到光学衍射极限的限制,普通光学显微镜难以胜任微米级以下颗粒物的测量,激光扫描共聚焦显微镜(confocal laser scanning microscope,CLSM)应运而生。CLSM主要用于观测荧光物质,又称为激光扫描共聚焦荧光显微镜。CLSM测量原理图是将被探测点位于成像透镜的焦平面,而共轭位置放置照明灯孔和探测针孔,因此被探测点所在位置为公共焦点。点照明可减小衍射影响,而探测针孔用于滤除焦平面以外的杂散光进入探测器,从而提升成像清晰度。具体测量原理如图 3所示,激光光束穿过照明针孔后形成点光源,依次经过二向色镜、物镜准确聚焦于样品的焦平面,照亮被荧光标记后的样品,其弹性反射光(波长等于入射光)会被分二向色镜镜阻挡,而荧光(波长大于入射光)通过二向色镜后成像于探测器。位于探测器前的探测针孔会阻挡被测样品附近非焦平面的荧光,只有焦平面(即被测样品像面)的荧光可以穿过探测针孔[16],达到清晰成像的效果。
CLSM主要用于荧光材料、组织切片、活体等的成像,还可以用于物质的定性、定量、定时分析等[17]。CLSM少用于粒径检测,粒径测量下限为200nm。对于亚微米级的颗粒物,可以估计出大小范围,但在精度上有所不足。应用有:氟化甲基丙烯酸酯颗粒(100nm~ 550nm)的粒径测量[18];结合大规模平行近似贝叶斯计算,估算聚苯乙烯颗粒(175nm~490nm)的大小[19]; 火山灰颗粒(5μm~54μm)成像检测[20]等。
表 1中列出了一些国外先进的激光共聚焦显微镜,分辨率在120nm左右,结合受激发射损耗成像技术可以达到20nm。

品牌 系列 横向分辨率/nm 奥林巴斯 FV3000 120 IXplore SpinSR 120 徕卡 TCS SP8 120 SR GSD 20 蔡司 LSM 980 120 尼康 N-STORM 接近20 N-SIM S 115 除光学显微镜外,电子显微镜、原子力显微镜(atomic force microscope, AFM)也是研究纳米颗粒物的常用显微镜。电子显微镜通过电子束撞击样品,使样品中的原子激发出次级电子,这些次级电子调制显像管得到反映样品结构的立体像。此方法可用于测量特征属性的分布,包括从电镜图像中的聚集体和团聚体中[21]或以单个颗粒形式存在的组成颗粒的最小外部尺寸。
原子力显微镜具有不受样品导电性的限制、可以在空气中使用、测量材料丰富等优点[22];还具有很高的分辨率,可以探测具有真实原子分辨率(0.1nm)的固体表面的结构和形态。测量误差通常在10nm以下,误差来源于探针接触样品造成的变形和样品配置过程中其它材料的残留,还需要用标准颗粒进行校准。广泛应用于纳米颗粒(聚苯乙烯颗粒、金、银[23]、SiO2[24]、TiO2、造影剂颗粒[25])的粒径测量。

光散射法是目前应用最为广泛的颗粒测量方法之一。颗粒的光散射特性与颗粒粒径密切相关,因此可以利用光的散射特性测量颗粒粒径[1]。光散射法主要有两类:静态光散射法和动态光散射法。其中静态光散射法包括角散射法、衍射散射法和光全散射法。
光散射法与其它方法相比有如下优势: (1)适用范围广。适用于固态、气态、液态以及部分两相流;(2)测量范围宽。可以测量3nm~1000μm的颗粒;(3)测量准确。误差小于1%~2%(标准颗粒);(4)测量重复性好。偏差小于1%~2%(标准颗粒)[26];(5)在线测量。光散射法测量时间短,已经应用于工业场合达到在线测量领域[8]。
光散射法的缺陷在于并非直接观测颗粒形貌,而是测量光学等效粒径,因此准确建立反演模型,往往决定了粒径分析精度。
显微镜法中往往需要较为复杂的样品沉积、制备要求,往往难以满足在线测量、或者具有活性颗粒物的粒径分析等场景。而光散射法在测量具有运动性、活性颗粒物具有独特优势。光散射法广泛应用于生物和生物制药领域中[27],如单个大分子溶质、蛋白质构象和胶体稳定性、非特异性蛋白质、蛋白质相互作用、稀溶液中可逆特定缔合、超分子组装等的表征。

(1) 角散射法。角散射法测量原理如图 4所示。聚焦的光束照射测量区中的待测颗粒,产生散射和吸收,由收集透镜和光电元件接收散射光通量,然后根据角散射理论反演颗粒粒径。目前常用的接收方式有两种:一种是接收多个角度的散射光通量; 另一种是接收一定角度范围的散射光通量。其中第2种应用更多。
根据角散射理论,当悬浮粒子系统被强度为I0的平行光束照射时, 粒子的折射率与周围介质的折射率不同,散射角光强度I(θ)与粒径分布f(D)可以通过第1类Fredholm积分方程联系起来,代入米氏散射函数可以写成:

(1) 式中,θ为散射角,ND是粒子弥散系的数密度;λ是入射光波长;D是球形颗粒物直径; Dmax和Dmin是粒子粒径积分上下限;f(D)为粒径分布函数;I1(θ, mλ, D, λ)和I2(θ, mλ, D, λ)是Mie散射函数,分别表示垂直和平行于散射面的散射光强度;mλ为粒子相对于周围介质的相对折射率。由(1)式即可反演颗粒的粒度分布。
角散射法的粒径测量范围为0.5μm~1000μm。测量误差小于10%,短测量波长和优化的测量角度选择有利于获得更好的测量结果。角散射法广泛应用于气溶胶、液溶胶的检测,如大气气溶胶[28]、LiCl气溶胶[29]、烟煤排放[30]、油品颗粒测量[31]等。在先验信息较少时,角散射法备受青睐,但是测量装置和粒径反演都比较复杂,也可能导致测量结果出现多个值,从而产生测量误差。除此之外,它对光学系统有较高的精度要求。
(2) 衍射散射法。衍射散射法通过测量前向小角度的散射光强反演颗粒粒径,又被称为小角前向散射法, 其原理图如图 5所示。激光器发出光束被针孔衰减和过滤,经准直和扩束后被样品池中的颗粒散射,并由傅里叶透镜成像到探测器阵列,根据颗粒的粒径范围预先在探测器上设计一系列的环,如图 6所示[32]。

环参数与粒径范围的关系是:

(2) 式中,m是环数,rm是第m个环半径,Dm是与第m个环半径相对应的粒径,f是透镜的焦距。
通过图像处理技术计算出每个环的散射光强度。对于在单次散射不相关的情况下的多分散粒子系统,探测器阵列上第i环(i < m)的散射光强度与粒度分布之间通过第1类Fredholm积分方程计算,可以表示为:

(3) 式中,m为探测器阵列上的环数; I1和I2为相对于散射平面的平行偏振光强度和垂直偏振光强度; Wi为权重分布; Di是每个子区间的平均粒径[Dm,Dm-1]; θm-1和θm是第m个环的内、外半径散射角。

(4) 式中, T为系数矩阵,E为m维探测能量向量,W为m维粒度分布向量。
衍射散射法的粒径测量范围约为0.5μm~1000μm[33],其测量误差可达4%,误差主要取决于折射率输入值与样品真实值不匹配和多次内反射,目前主要用于气溶胶、液溶胶监测, 如蛋白质-溶菌酶[34]、细胞[35]、悬浮等离子喷涂[36]等。近几年的研究中,大都是对反演算法进行改进,以提高反演精度,如Tikhonov正则化[32, 37]、Chin-Shifrin积分变换[38]、迭代非负Philips-Twomey(iterative non-negative Philips-Twomey, INNPT)算法[39]、对偶积分反演[40]、人工鱼群算法[41]等。
(3) 光全散射法。光全散射法又被称为光谱消光法, 其测量原理如图 7所示。它基于Lambert-Beer定律,当一束光强为I0的平行单色光,穿过颗粒物悬浮液会发生散射和吸收导致透射光的衰减。如果忽略多重散射(颗粒间距离大于3倍粒径)和相互作用的影响,将入射光强与透射光强之比I(λi)/I0(λi)定义为消光比,可由下式计算:

(5) 式中,λi(i=1,2,3, …, k)为入射光波长,通过实验测量获得,在反演过程中作为已知参数; L是光程,nD是悬浮液中的颗粒物总数; Qext(λi, mλ, D)是单个球形颗粒的消光效率,它是波长λi、粒径D和相对折射率mλ的复函数。由(5)式即可反演粒径分布f(D)。
光全散射法的测量范围为50nm~10μm,它的测量误差主要取决于颗粒浓度,低浓度高透射率容易产生噪声,高浓度则会出现多重散射效应。光全散射法具有测量速度快、重复性好、原理简单、操作简便、对仪器设备要求不高、还能测量颗粒浓度等优点。消光法广泛应用于气溶胶(如大气气溶胶[42-43]、石英气溶胶[44])、液溶胶(微藻细胞[45]、血红蛋白[46])的监测,尤其是对于大气气溶胶的的遥感监测具有独特的优势。由于各种反演算法的性能还不能令人满意,因此,在近几年的研究中,两种算法结合以提高测量精度和重复性的方式受到研究者们的青睐,如人工蜂群算法与模式搜索[47]、Tikhonov正则化与平均法[48]、Tikhonov正则化与主成分分析[45]等。

动态光散射(dynamic light scattering, DLS)又称光子相关光谱(photon correlation spectroscopy, PCS),它是基于布朗运动和Mie散射的一种测量方法。
测量原理如图 8所示。激光器发出光束经准直后照射含有待测颗粒的样品槽,诱导颗粒产生极化,振动的电偶极子作为次波源向各个方向辐射电磁波(散射光),由于颗粒的布朗运动,偶极子的位置也不断变化,散射光强信号会产生起伏,由布朗运动的基本原理可知,颗粒粒径越小,运动越剧烈,因此散射光的起伏也会更加剧烈。动态光散射通过散射光起伏与颗粒粒径的关系来反演颗粒的粒度分布。
利用光子计数器和数字相关器在某个角度接收散射光并计算自相关函数。其中,电场自相关函数g(1)(τ)为:

(8) 式中,Q为散射矢量的模,DΓ为粒子的扩散系数,kB为玻尔兹曼常数,T为绝对温度,η为溶液的粘度,d为样品的粒径。kB, T和η为已知量。
结合(6)式~(8)式,就建立了自相关函数与颗粒粒径的关系,可以通过测量计算出自相关函数,进而反演颗粒粒径。
动态光散射法测量范围约为3nm~3μm。它的测量误差主要来源于测量角度的选择和反演算法的性能,它在测量前向角度和反演单峰分布时均具有较高的准确性,可达到5%以下;在反演多峰分布时误差较大,已有研究表明,相对于单角度动态散射光系统,多角度动态光散射提供了重复性好和更准确的粒径与粒度分布,双峰分布的反演误差可达6%[49]。
动态光散射法样本制备过程简单,可用于干性、湿性颗粒物粒径分布测量,在估算多分散系,多组分纳米粒子尺寸的应用中,动态光散射法输出粒径分布而非平均粒径[50]。动态光散射法是光散射法中目前应用最广的一种,被广泛应用生物医学、工业监测等领域,例如金[50]、银[51]、氧化锌[52]、药物佐剂[53]、淀粉脂肪酸混合物[54]、蛋白质聚集[55]、纳米气泡[56]和灰尘[57]等的监测。国际上知名的动态光散射仪包括英国马尔文公司ZetaSizer系列,美国布鲁克海文仪器公司Nano DLS系列,可达到1nm的测量精度。但是,动态光散射法测量粒径在实际应用中同样存在一些问题。对单分散颗粒样品,可以通过拟合动态光散射实验数据的相关曲线求出散射谱线宽,直接求出颗粒粒径。但对于多分散颗粒系,情况则复杂得多,通过求解第1类Fredholm积分方程反演颗粒粒径,这一求解过程属于非适定性问题,任何微小的数据扰动都可能造成巨大偏差,需要合适的反演算法才能得到接近理论粒径分布的解。为了提高粒径反演的准确度,不断有学者提出多角度光散射系统设计和粒径反演算法,如迭代递归非负Tikhonov-Phillips-Twomey(iterative recursion nonnegative Tikhonov-Phillips-Twomey,IRNNT-PT)算法[58]、多重惩罚加权正则化[59]、加权贝叶斯反演算法[60]、小波迭代递归非负Tikhonov-Phillips-Twomey(wavelet iterative recursion nonnegative Tikhonov-Phillips-Twomey,WIRNNT-PT)算法[61],这是目前的研究热点。另外,颗粒形貌差异也会带来粒径反演结果的偏差,研究非球形颗粒物粒径反演算法亦是目前的重要研究方向[62]。

激光全息测量法基于洛伦兹-米氏(Lorenz-Mie)光散射理论,测量原理如图 9所示。激光器发出具有高度相干性的光束,依次经过扩束镜和分光镜,其中一束穿过含有待测颗粒的测量区,另一束光波不经过测量区[32]。两束光波在图像传感器处产生干涉,并由用图像传感器(如电荷耦合元件(charge-coupled device, CCD)或互补金属氧化物半导体(complementary metal oxide semiconductor, CMOS))采集得到全息图,基于瑞利-索末菲衍射理论和角谱理论,可将全息图重建为粒子场的3维分布,然后用阈值分割等算法计算颗粒粒径。
将全息图重建为粒子场3维分布的方法有:菲涅耳积分算法、角谱算法、卷积算法[63]。以菲涅耳积分算法为例:菲涅耳积分算法基于瑞利-索末菲衍射理论,当衍射距离即全息图平面到衍射平面的距离能够满足菲涅耳近似条件时,菲涅耳衍射公式可以简化表示为傅里叶变换的形式:

(9) 式中,U(x, y)为模拟衍射的再现物象,U(x0, y0)为图像传感器记录下来的全息图光强分布与再现光波的乘积。使用菲涅耳积分变换算法进行全息图像重建只需要进行一次傅里叶变换就可以得到再现物像,得到的物像经过阈值分割可以提取出被测颗粒粒径大小和分布情况等信息。
全息成像的记录光路有同轴光路和离轴光路两种,以全息图中心、待测物体中心的和参考光中心是否在一条轴线上为标准;同轴全息光路对系统的稳定性和光源的相干性要求不高,而离轴全息光路对系统光源的相干性要求较高,因此采用同轴全息光路具有较大的优势[63]。
激光全息测量法是一种3维测量方法,测量下限为2μm,相对误差一般小于6%;它的分辨率受像素大小的限制,测量误差还来源于环境干扰、相机中的帧丢失以及激光器的波长波动。激光全息测量法在水中污染物区分[64]、生物学中细胞培养[65]、病毒监测[66]、工业中液滴[67]、云滴[68]、金属液滴[69]、煤粉[70-72]、微生物颜料[73]等的监测中已有应用,可以提供颗粒的粒度分布、速度、数目、运动轨迹等信息, 其缺点是设备昂贵,系统结构较复杂,对测量环境要求高,数据计算较复杂。采用深度学习神经网络替代传统重建算法直接重建出粒子场,可提高计算效率[74]。在众多的全息系统中,数字无透镜全息以结构简单、成本低廉、灵敏度高、可消除噪声和像差等优点而备受青睐。

纳米颗粒跟踪分析法(nanoparticle tracking analysis, NTA)基于布朗运动和Stokes-Einstein原理, 它的测量过程如图 10所示。激光束穿过含有待测颗粒的样本室,利用装有摄像头的长工作距离显微镜接收散射光,拍摄布朗运动下移动颗粒的视频文件。
NTA测量粒径的原理是溶液中的粒子会经历恒定的随机运动。单个粒子的移动取决于它们的大小,其中小粒子的移动速度比大粒子快。根据Stokes-Einstein方程,在一定温度下悬浮于已知粘度的液体中的球形颗粒由于布朗运动而产生的速度与颗粒直径成正比[52]。由方程计算出样品粒径。同时,通过仪器内置软件,对观察到的每一个颗粒进行跟踪分析,得到粒度分布以及颗粒物浓度。
NTA法不会破坏样本,保证了测量数据的真实性和有效性,它的测量偏差约为6%,误差主要来源于颗粒非布朗运动分量,这将造成偏置误差,往往需要修正。同时具有荧光增强检测能力,标记检测特定样本,免受复杂样本环境的影响。无需预先知道质量、折光度和颗粒材料的流体力学直径[75],并且能对悬浮液中粒径分布范围较宽(10nm~2000nm)的颗粒进行全方位表征。缺点是该方法适用于固体和球形颗粒,对于大分子或棒状颗粒,计算出的是流体动力学直径,而不是真实粒径。NTA法广泛用于蛋白质聚集体[76-77]、细胞外囊泡[78]、病毒[79]和药物输送载体[80-81]、废水监测[82]、生物制药[83]以及环境修复[84]等领域。

将显微镜法、光散射法和激光成像法3类方法的测量范围以及适用对象总结于表 2中,并补充了电子显微镜和原子力显微镜的粒径测量范围进行对比。可见, 除了普通股光学显微镜和衍射散射法主要用于干性、粉体颗粒物,其它方法均可用于粉体和液体溶胶两种性质的颗粒物粒径检测。表 3中总结了各类方法的优缺点。

摘要: 粒径测量技术广泛应用于能源、材料、医药、化工、冶金、电子、机械、轻工、建筑等诸多领域, 对于改善产品质量、提高生产效率、降低能源消耗、实现精准医疗、监测环境污染等具有十分重要的意义。基于光学原理的粒径测量方法具有响应速度快、粒径测量范围可达纳米级等优势, 但也存在一些问题, 需要不断完善。总结了基于光学原理的3类粒径测量方法, 即显微镜法、光散射法、激光成像法; 介绍了各种方法的测量原理和应用领域, 对比分析了不同粒径测量方法的测量范围和优缺点。在此基础上对粒径测量方法的发展方向进行了展望。Abstract: Particle size measurement technology is widely used in many fields such as energy, materials, medicine, chemical industry, metallurgy, electronics, machinery, light industry, construction, etc., and has a very important meaning for improving product quality, increasing production efficiency, reducing energy consumption, realizing precision medicine, monitoring environmental pollution, etc. The particle size measurement method based on the optical principle has the advantages of fast response speed, and the particle size measurement range can reach nanometer level. However, there are still some problems in practical applications and need to be continuously improved. Three types of particle size measurement methods based on optical principles: Microscopy, light scattering, and laser imaging were summarized in this paper, including the measurement principles, measurement ranges, advantages and disadvantages, and application areas of various methods.
